Description
R2010
Highlights
High Quality: Extract intact high quality total RNA (including small/micro RNAs) from Gram-negative/postive bacteria, yeast, and fungi
Homogenization: Ultra-high density BashingBead technology for complete disruption and sample lysis.
Inhibitor-Free: RNA is ready for all downstream applications including sequencing, RT-PCR, microarray, etc.
Description
The Quick-RNA Fungal/Bacterial Microprep Kit is designed for the simple and rapid isolation of RNA from tough-to-lyse fungi, including A. fumigatus, C. albicans, N. crassa, S. cerevisiae, S. pombe, as well as Gram (+/-) bacteria, algae, and protozoa. The procedure is easy and can be completed in minutes: fungal and/or bacterial samples are rapidly and efficiently lysed with our state of the art, ultra-high density BashingBeads. Zymo-Spin column technology is then used to isolate the RNA that is ideal for downstream molecular-based applications including Next-Gen sequencing, RT-PCR, microarray, hybridization, etc.

‘CMC was purchased from Sigma (St. Louis, MO).. Total RNA was isolated from cells grown in TSB without pectin and in TSB supplemented with 0.4% w/v of either PGA, apple pectin, or RG-I from soy at 12.75 h after inoculation with the ZR Fungal/Bacterial RNA Miniprep kit (Zymo Research, Irvine, CA).. Ten µg of RNA was treated with DNase using the TURBO DNA-free Kit (Thermo Fisher, Waltham, MA).’

‘of the gene were concatenated and a multiple alignment was constructed using BioEdit Version 7.2.5 ( ) to determine nucleotide and amino acid changes. . Total RNA was isolated from DIF-free 14-day-old PDA plates using a ZR Fungal/Bacterial RNA MiniPrep Kit (Zymo Research, Irvine, CA, United States) according to the supplier’s instructions.. All RNA was analyzed for quantity and quality spectroscopically on 1% TBE agarose gel.’

(2017), for whole genome tiled amplicon sequencing using a novel protocol, based on the method of Quick et al. developed for Zika virus sequencing. . Prior to transporting the samples, an equal part of DNA/RNA ShieldTM 2X Concentrate (Zymo Research) was added to each mosquito pool to stabilize the RNA and inactivate WNV.. The pools consisted of mosquitos in approximately 1.3 ml of TE buffer (Invitrogen, AM9858).
